Carcinogenesis in Swiss mice by isonicotinic acid hydrazide.

چکیده
There have been several demonstrations of the limited tumorigenicity of INH in mice. The 1st such experiment was re ported by Juhász et al. (3, 4), who reported the induction of pulmonary adenomas, lymphomas, and leukemia.s in randomly bred mice injected i.p. with INH. In a subsequent study these same investigators found only an induction in lymphomas and leukemias but reported the occurrence of only 1 pulmonary adenoma (5). Other workers have reported on the induction of pulmonary adenomas (1, 7, 8, 10, 11) and it has more recently been suggested that this action may be mediated through hypo thetical metabolism of INH to hydrazine sulfate, which has also been shown to induce lung adenomas (2) in BALB/c mice. In 1 study in the rat (9) no tumors were induced with repeated injec tions of INH and, in fact, in another study in this species the production of hepatomas by p-dimethylaminoazobenzene was inhibited by combined administration of this carcinogen with INH (6). The clinical implications of these various findings have been discussed and many divergent viewpoints presented. The present study has been undertaken because of the clinical interest in this situation. Prior studies reported were terminated before the end of the life-span of the mice. It was felt that addi tional information might be obtained if the experiments were allowed to run to their conclusion.

Nicotinamide-adenine Dinucleotide-glycohydrolase Activity in Experimental Tuberculosis.
1. The specific NAD-glycohydrolase activity is increased 70 and 50% over the normal in lung and liver tissues respectively of tuberculous mice. 2. Concomitant with the increase in the NAD-glycohydrolase activity, the NAD-isonicotinic acid hydrazide-exchange activity also is increased in infection. The isonicotinic acid hydrazide analogue of NAD formed by the lung enzyme from tuberculous mice ha...

Eight new complexes of Cu(II), Co(II), Ni(II) and Zn(II) with isonicotinic acid hydrazide (isoniazid, (INH)) and isonicotinic acid (1-naphthylmethylene)hydrazide (INNMH), having the formula of the type [M(INH)(ac)2] or [M(INNMH)(ac)2] (M = Co(II), Ni(II) and Zn(II)) and [Cu(INH)(ac)2]2, [Cu(INNMH)(ac)2]2, were synthesized and characterized. All complexes were characterized based on elemental an...
متن کاملCharge Transfer Interactions between Isonicotinic Acid Hydrazide and Cupric Ions
The interaction of Cu2+ with isonicotinic acid hydrazide (INH) and some of its derivatives has been investigated by means of electron spin resonance, nuclear magnetic resonance, and optical absorption studies. It could be shown that an interaction exists between the metal ions and the hydrazine side chain in addition to the very weak interaction with the ring nitrogen. The results obtained indi...

A procedure for blood plasma is presented in which the test compound is converted ...
متن کاملPeroxidase-mediated oxidation of isoniazid.
Oxidation of isonicotinic acid hydrazide (isoniazid) by horseradish peroxidase at the expense of H2O2 yielded reactive species which were able to reduce nitroblue tetrazolium and bleach p-nitrosodimethylaniline. Nicotinic acid hydrazide oxidation did not cause these effects. At slightly alkaline pH, oxidation of isonicotinic acid hydrazide by horseradish peroxidase proceeded at the expense of m...
